Developmental Therapeutics in Multiple Myeloma and Hematological Malignancies

There is a worldwide rising incidence and prelavence of hematological malignancies. The past few years have witnessed a huge advancement in the knowledge of genetic and molecular events supporting the pathogenesis of leukemias, multiple myeloma and Waldenstrom's macroglobulinemia. On these achievements, several new therapeutics have been rapidly translated to the clinical setting, with a deep impact in the management of these disease, leading to a paradigm change in the clinical practice. However, even if presently available treatments have produced benefits in terms of long-term survival, the cure of most of these malignacies remains an unmet need.

Innovative research on hematopoietic malignancies for development of rational therapeutic approaches must rely on relevant basic issues including the tumor dependency by the non-tumor compartment of the bone marrow milieu and the emerging role of important regulatory intra- and inter-cellular newtworks like non-coding microRNAs. Recently, our research programs have been based on these two important fields of investigation:

a) the setting of innovative murine models for the recapitulation of the human bone marrow milieu in vivo (SCID-hu, SCID-synth-hu) which allow the experimental validation of new agents targeting not only tumor cells but also the tumor microenvironment. In this specific area of investigation and technological advancement, we provided the scientific accademic and corporate community with robust and reliable systems for preclinical selection of investigational therapeutics with predictive potential for clinical translation. Some of these compounds which have been developed by the use of our in vivo models have already reached the IND status and are presently in phase I/II clinical trials.

b) the development of innovative anti-tumor agents derived from the study of the microRNA network. Our research efforts recently led to the selection of synthetic miRNA mimics or miRNA inhibitors which demonstrated a reliable anti-tumor effectiveness in vivo. Notably, this activity has been demonstrated to rely on validated target inhibition, providing evidence of high selective molecular activity. This research efforts, which also involves the development of original nanodelivery systems and original molecule design, has been strongly supported by the Italian Association for Cancer Reseach (AIRC), Special Program Molecular Clinical Oncology - 5 per mille" n. 9980, 2010/15, which specifically focus on multiple myeloma and B-cell chronic lymphocytic leukemia and is presently ongoing.

Pierfrancesco Tassone, M.D. is serving as Adjunct Professor at Temple University's College of Science & Technology, Philadelphia, PA. After MD graduation in Italy, his scientific career begun as Research fellow at New York Medical College, Valhalla, NY, and subsequently he has been appointed as Research Associate at University of California San Diego, CA, and at Dana-Farber Cancer Institute and Harvard Medical School, Boston MA. Presently, Dr. Tassone is Full Professor of Medical Oncology at the Magna Graecia University of Catanzaro, and Director of the Medical Oncology Unit at Tommaso Campanella Cancer Center. Dr. Tassone is PI of the Research Network in the Special Program Molecular Clinical Oncology: A research platform for miRNA-based treatment of multiple myeloma and B-cell chronic lymphocytic leukemia - 5 per mille" n. 9980, 2010/15, funded by the Italian Association for Cancer Reseach (AIRC).
